Services
MAb development
- Procurement of mice
- Mouse and Rat immunization protocols for whole cells, purified proteins, peptides and other antigens
- Antibodies for IHC, WB, IP, functional, blocking, agonist, anti-idiotype. For in vitro, ex vivo, or in vivo experiments, and as proof of concepts for potential therapeutic use
- Antigen sources: human, mouse, elephant, dog, fungus, and other proteins from other species
- Use of regular mice, KO mice, or "Humice" (to generate fully human antibodies upon recombinant construction)
- Testing mouse/rat sera for positive response
Fusion and Hybridoma Screening
- Cell Fusion of immunized mouse/rat spleen or lymph node cells with drug-sensitive myeloma fusion partner for generation of hybridomas
- Growing and isolation of primary hybrid fused cells by tissue culture
- Screening by ELISA
- Provide investigators with culture supernatants for antibody assays and validation with their own protocols
- Special screening by FACS in collaboration with Flow Cytometry Lab South Campus (Karen Ramirez, kmramirez@mdanderson.org)
- Special screening for immunohistochemistry antibodies in collaboration with the Histology Core (Dr. Gabriela Raso, graso@mdanderson.org)
Cloning
- Single cell cloning of desired hybridomas to create a stable cell line
- Freezing representative hybridoma cell lines for long term viable storage in our liquid nitrogen tanks at no extra charge
Small-scale antibody production
- Production of antibody-containing culture supernatants (2 mLs), to provide the Core user with all the positive clones for further analysis and selection in their labs
- We support our users all the way through the selection providing all results to users
Antibody purification
- Small/medium scale production of purified monoclonal antibodies from custom hybridoma supernatants or hybridomas stocked in the MAF's extensive library
- Protein A/ Protein G affinity chromatography purification
- Affinity Purification of monoclonal antibodies using FPLC (Fast Protein Liquid Chromatography)
- Purification from mice ascites fluid
- In vitro purification from non-murine hybridomas, recombinant CHO and/or 293-HEK cells
- Large scale purification of antibodies for in vivo experiments (information provided to contact the recommended company)
Antibody characterization
- Murine Isotyping
